KYPROLIS and Nplate, $2.9 billion of oncology sales, lose key American patents in 2027 and 2028.
Amgen's oncology growth is carried by BLINCYTO and IMDELLTRA, but half of the band's sales come from older drugs whose protection is ending. KYPROLIS, for multiple myeloma, has United States compound patents expiring in December 2027, with pediatric exclusivity to June 20281. Nplate, for low platelet counts, has a formulation patent expiring in February 20282.
Those two drugs are large. KYPROLIS sold $1,412 million and Nplate $1,524 million in 20253. KYPROLIS is already shrinking, down about 6% from $1,503 million in 20244, and $314 million in the second quarter of 20265. Its competitors, according to the 10-K, include DARZALEX and CARVYKTI from Johnson & Johnson and POMALYST and REVLIMID6, a field that has moved on to newer mechanisms.
KYPROLIS is a small molecule, a proteasome inhibitor. When its patent ends, generic copies can arrive quickly, as Amgen saw with Sensipar7. Nplate is a biologic, so a copy takes longer, but the formulation patent is a thinner shield than a composition patent.
Together the two sold $2,936 million in 20258, about 44% of the oncology band's $6,660 million9. BLINCYTO and IMDELLTRA sold $2,186 million10.
The test is whether the new drugs outgrow the loss of the old ones. By 2028 BLINCYTO and IMDELLTRA need to exceed the $2.9 billion KYPROLIS and Nplate sold in 2025; if they do not, Amgen's oncology line will shrink in 2028 and 2029 despite its successes.
