Amgen (AMGN) — threat to the moat
Prolia and XGEVA, $6.5 billion of sales in 2025, lost their patents and fell by a third in the second quarter of 2026.
Denosumab was Amgen's largest franchise, and its erosion is the defining event of 2026. Prolia, for osteoporosis, and XGEVA, for bone complications of cancer, contain the same active ingredient1. Together they sold $6,498 million in 20252, about 17.7% of total revenues3. Their key American patents on the RANKL antibodies expired in February 2025, and select European ones in November 20254.
The copies have come quickly. In the second quarter of 2026 Prolia's sales fell 32% to $759 million, driven by 20% lower volume and 12% lower net selling price as multiple biosimilars have launched globally with more biosimilars expected5. XGEVA fell 34% to $352 million, on 22% lower volume and 8% lower net selling price6. Prolia's American sales fell from $745 million to $478 million7. In the first half the pair sold $2,249 million, against $3,319 million a year earlier8.
Amgen has seen this before. Neulasta sold $4,534 million in 2017 and $435 million in 2025 after its copies arrived910, and its 10-K says that once several biosimilars launch, competition intensifies rapidly, resulting in accelerated net price declines for both the reference and the biosimilar products11.
The difference now is that Amgen's growth drivers are large enough to absorb it. Total revenues still rose 10% in the second quarter12.
Prolia and XGEVA are one molecule sold two ways. The 10-K explains that they contain the same active ingredient but are approved for different indications, patient populations, dose and frequency of administration13. Prolia was launched in the United States and Europe in 201014, grew every year from $1,968 million in 201715 to a peak of $4,414 million in 202516. The first quarter of 2026 showed how fast the turn came: Prolia sold $727 million and XGEVA $411 million17.
The threat is sized by how far the pair falls. If denosumab sales in 2026 come in near $4.5 billion, a decline of about a third, the growth drivers can carry it; if the decline accelerates toward the Neulasta pattern, about 90% over eight years, Amgen loses a sixth of its revenue in a few years.
