Amgen (AMGN) — threat to the moat

In 2026 Amgen handed rocatinlimab back, dropped bemarituzumab in gastric cancer and stopped AMG 513, while its research budget rose 52% in two years.

The acquisitions that worked are visible in the sales tables. The ones that did not are in the footnotes, and 2026 produced several. In January Amgen and Kyowa Kirin agreed to terminate the rocatinlimab collaboration and to transition control of the global development and commercialization program to Kyowa Kirin1. Rocatinlimab was Amgen's candidate for atopic dermatitis, a large market.

In February the 10-K said Amgen does not intend to pursue regulatory approval of bemarituzumab in first-line gastric cancer2. In the second-quarter 2026 release it said future development of AMG 513, an obesity candidate, will be discontinued3.

Stopping failing programmes is good discipline, and every drug maker does it. The concern is what it says about the pipeline's yield. Amgen spent $7,272 million on research and development in 2025, up about 52% from $4,784 million in 202345, about 19.8% of revenue6, and three visible programmes ended within months.

Leadership is also changing. David Reese, the chief technology officer, is retiring7, and Peter Griffith is being succeeded as chief financial officer by Thomas Dittrich from 1 September 20268. The chief executive, Robert Bradway, has led the company since 20129.

The figure to watch is how many late-stage programmes reach filing. If the next two years produce more terminations than approvals, while research spending keeps rising, Amgen will be spending more to find less and will lean further on buying drugs from others.
